Human Neural Stem Cells from Gentaur are differentiated from a human induced pluripotent stem cell line (hiPSC), which is generated using mRNA reprogramming technology from Human Dermal Fibroblasts (HDF). The monolayer HDF-hiPSC are efficiently converted to neural epithelium using HPSC Neural Induction Medium, a serum-free medium for rapid and efficient neural induction of human pluripotent stem cells (hPSCs). Synergistic inhibition of glycogen synthase kinase 3 (GSK3) and transforming growth factor b (TGF-b) differentiates HDF-hiPSCs to homogenous neural stem cells (NSC) in the presence of human leukemia inhibitory factor (LIF) within 7 days.The derived NSC are characterized by immunofluorescence with antibodies specific to nestin and SOX2. The cell population is highly pure: andgt;90% of cells express Nestin and andgt;80% of cells are SOX2 positive. After reviving, NSC can be maintained in HPSC Neural Induction Medium as an adherent culture or form neural spheres in EGF and bFGF containing neural stem cell medium. The cells are able to form rosette and neural tube-like structures after being passaged and plated at high density on Matrigel coated culture vessels in neural stem medium containing EGF and bFGF. HiPSC-NSC can be further expanded for months as a monolayer on MatrigelTM coated culture vessels or in suspension as neural spheres when seeded on ultra-low binding culture vessels in neural stem cell medium. NSC are multipotent and able to differentiate into various neuronal and glial subtypes. Specific patterning cues, such as SHH, retinoic acid and FGF8, can be added after reviving to direct the cells to different neural lineages.HiPSC-NSC are cryopreserved at passage 0 and delivered frozen. Each vial contains andgt;1 x 10^6 cells in 1 ml volume. Cells are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi.

Gentaur Human iPSC-Derived Neural Stem Cells (NSCs) are derived from integration-free, induced pluripotent stem cells (iPSCs) under fully defined neural induction conditions. The NSCs express typical markers of cerebral cortical neural stem and progenitor cells such as PAX6, FOXG1 and nestin, and spontaneously form polarized neural tube-like rosette structures when plated as a monolayer in culture (see below). Additionally, Gentaur NSCs are capable of generating a spectrum of cerebral cortical excitatory and inhibitory neurons that are electrically active and have the ability to form functional synapses and circuits in vitro. After thawing and plating, the neural stem cells terminally differentiate to acquire mature electrophysiological properties, and form functional synaptic networks over a period of 40 ~ 50 days. Gentaur NSCs are easy to differentiate to neurons or mixed neural cell types, following our protocols and using our tailored media and reagent bundles. A highly pure population of neurons can be generated from Gentaur NSCs following the synchronous differentiation protocol. Using our specialized coating reagents, neurons derived from Gentaur NSCs can be maintained in culture long-term (andgt;1 year). Human Human IPSC-Derived Neural Stem Cells that have been genetically edited using CRISPR-Cas9 technology to introduce the R406W mutation (CGGandgt;TGG) into the MAPT gene. This line is homozygous for the R406W mutation so both alleles contain the mutation. The R406W mutation in MAPT has been implicated in familial frontotemporal dementia and parkinsonism (Hutton et al., 1998; Behnam et al., 2015) and has been seen in patients with clinical presentation resembling Alzheimer's disease (Rademakers et al., 2003; Lindquist et al., 2008). The R406W mutation reduces the ability of MAPT/tau to bind to microtubules in vitro (Hong et al., 1998).

RFP-HBMMSCs are selected from puromycin resistant HBMMSCs after transfected with RFP expressing lentiviral particles. RFP-HBMMSCs are supplied in cryovials at passage 3-4. Each lot of RFP-HBMMSCs is tested to ensure the cells can be passaged at least for three times (i.e., approximately 9 to 10 population doublings) after thaw. RFP-HBMMSCs should be maintained in complete growth media (Mesenchymal Stem Cell Growth Medium (MSCGM)) following the detailed protocol described below.
