BKS db Mouse Retinal Microvascular Endothelial Cells are isolated from the retinal microvascular of Mice homozygous for the diabetes spontaneous mutation (Lepr/db) manifest morbid obesity, chronic hyperglycemia, pancreatic beta cell atrophy and become hypoRIemic. BKS db Mouse Retinal Microvascular Endothelial Cells are grown in T25 tissue culture flasks pre-coated with gelatin-based solution for 2 min and incubated in Gentaur's Culture Complete Growth Medium generally for 3-7 days. Cultures are then expanded. Prior to shipping, cells at passage 3 are detached from flasks and immediately cryopreserved in vials. Each vial contains at least 0.5×106 cells per ml. The method we use to isolate primary endothelial cells was developed based on a combination of established and our proprietary methods. These cells are pre-coated with PECAM-1 antibody, following the application of magnetic beads pre-coated with secondary antibody.

mBMECs were isolated from E18 embryonic brain cortex microvessels of Balb/C mouse and transformed by transfected with SV40 middle T-antigen. GFP-mBMECs were selected puromycin resistant mBMECs from GFP-expressing lentiviral particle. The cells are shipped in frozen vials(the cells are provided @ passage 6) transfected mAECs. The cells are shipped in proliferating culture with 90 confluence (the cells are provided @ passage 4-6). DMEM containing 20% FBS is recommended for cell culture.

Mouse Liver Microvascular Endothelial cells (mLivMECs) are isolated from livers of 3-week old male C57BL/6 mice and GFP expressing mLivMECs-GFP were selected from puromycin resistant cells after transfected with lentiviral expressing GFP. Cells are shipped in a frozen vial (provided @ passage 3, with 5 x 105cells/vial). mLivMECs-GFP are cultured with specially formulated Endothelial Growth Medium. Since the cells have limited proliferation capacity (cells may be expanded for a maximum of 2-3 passages) in vitro, long term culture of mLivMECs is not recommended, even cultured following the detailed protocol described below).
