Fibroblasts are mesenchymal cells derived from the embryonic mesoderm. They have been extensively used for a wide range of cellular and molecular studies. This is mainly because they are one of easiest types of cells to grow in culture, and their durability makes them amenable to a wide variety of manipulations ranging from studies employing gene transfection to microinjection. Fibroblasts within tissues are exposed to a dynamic mechnical environment, which influences the structure integrity of both healthy and healing soft tissue. They secrete a non-rigid extracellular matrix that is rich in type I and/or type III collagen. There is good evidence that fibroblasts in different parts of the body are intrinsically different and display different regulatory functions on surrounding cells. For example, splenic fibroblasts have been shown to regulate the development of NK cells from blood progenitors through an expression of bioactive IL-15. HSF from Gentaur Research Laboratories are isolated from human spleen tissue. HSF are cryopreserved at primary culture and delivered frozen. Each vial contains 5×10^5 cells in 1 ml volume. HSF are characterized by their spindle morphology and immunofluorescent method with antibody to fibronectin. HSF are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast and fungi. HSF are guaranteed to further expand for 15 population doublings at the condition provided by Gentaur Research Laboratories.

BALB/c Mouse Primary Lymphatic Fibroblasts from Gentaur are isolated from tissue of pathogen-free laboratory mice. Mouse Primary Lymphatic Fibroblasts are grown in T25 tissue culture flasks pre-coated with gelatin-based solution for 0.5 hour and incubated in Gentaur Culture Complete Growth Medium generally for 3-7 days. Cultures are then expanded. Prior to shipping, cells are detached from flasks and immediately cryo-preserved in vials. Each vial contains at least 1x10^6 cells per ml and is delivered frozen.Mouse Primary Lymphatic Fibroblasts are negative for bacteria, yeast, fungi, and mycoplasma. Cells are tested for expression of marker using the antibody of anti-FSP1/S100A4 by immunofluorescence staining. Cells can be expanded for 3-5 passages at a split ratio of 1:2 under the cell culture conditions specified by Gentaur. Repeated freezing and thawing of cells is not recommended.Standard biochemical procedures performed with cell cultures include the assay of cell to cell interaction, RT-PCR, Western blotting, immunoprecipitation, immunofluorescent staining, flow cytometry or generating cell derivatives for desired research applications.
