Human Tracheal Smooth Muscle Cells

Smooth muscle tissue is found in the tunica media layer of large and small blood vessels and in the walls of hollow organs like the bladder and the uterus. Smooth Muscle Cells (SMC) possess all the same types of filaments, but, depending on the tissue of origin, differ significantly in mechanical and physiological properties. Therefore, Gentaur offers a range of Human Smooth Muscle Cells produced at Gentaur's cell culture facility from normal human tissue of different origins. The cells are isolated from the aorta, the coronary artery, the pulmonary artery, the umbilical artery,the trachea, the bronchi, the bladder,the prostate, and the uterus. Shortly after isolation, all Gentaur Human Smooth Muscle Cells are cryopreserved at passage 2 (P2) using Gentaur's proprietary, serum-free freezing medium, Cryo-SFM. Each cryo vial contains more than 500,000 viable cells after thawing.Proliferating cell cultures are made from 500,000 cryopreserved cells that have been thawed and cultured for three days at Gentaur.

Rat Tracheal Smooth Muscle Cells

Rat Tracheal Smooth Muscle Cells from Gentaur are isolated from tissue of Sprague-Dawley Rats. Rat Tracheal Smooth Muscle Cells are grown in T75 tissue culture flasks pre-coated with gelatin-based coating solution for 0.5 hour and incubated in Gentaur's Cell Culture Medium for 3-7 days. Cultures are then expanded. Prior to shipping, cells are detached from flasks and immediately cryo-preserved in vials. Each vial contains at least 1x10^6 cells per ml and is delivered frozen.Cells are negative for bacteria, yeast, fungi, and mycoplasma and are characterized by immunofluorescent staining with antibodies to α-smooth muscle actin. Cells can be expanded on a multiwell culture plate ready for experiments under the cell culture conditions specified by Gentaur.Repeated freezing and thawing of cells is not recommended.Standard biochemical procedures performed with smooth muscle cell cultures include the assays of cell to cell interaction, PCR, Western blotting, immunoprecipitation, immunofluorescent staining, immunofluorescent flow cytometry or generating cell derivatives for desired research applications.

Human Esophageal Smooth Muscle Cells

Smooth muscle is responsible for the contractility of hollow organs, such as blood vessels, the gastrointestinal tract, the bladder, and the uterus. Its structure differs greatly from that of skeletal muscle. The human esophagus contains three layers of muscle in its walls, the outer longitudinal and inner circular layers of the main muscular coat and the muscular layer of the mucosa. Visceral smooth muscle cells makes up all three layers along the entire organ except for the most rostral few centimeters. Smooth muscle contraction is critical to peristalsis in the human esophagus and the contraction may be mediated by activation of phospholipase through two distinct mechanisms (increased intracellular Ca2+ and G protein activation) and activating PKCepsilon-dependent mechanisms. The human stomach contains three layers of muscle in its walls, the outer longitudinal, the middle circular and the inner oblique and visceral smooth muscle cells makes up all three layers along the entire organ. Human Intestinal Smooth Muscle Cells

Smooth muscle contraction is the fundamental event in gastrointestinal motion. Although many of the biochemical mechanisms underlying the excitation-contraction coupling are not yet defined, it is known that cytosolic Ca2+ is the essential component in the coupling phenomenon. Inflammation of the human intestine causes thickening of the smooth muscle layers which results from the increases in the smooth muscle-specific actins. The increased smooth muscle actins may affect force production and further demonstrate the plasticity of smooth muscle in the inflamed intestine. Studies also show that human intestinal smooth muscle cells respond to IL-1beta and TNF-alpha stimulation by releasing IL-6, which might significantly contribute to the overall systemic inflammatory response.
